研究内容:
恶性肿瘤本质上是由体细胞DNA变异诱发的系统疾病,本实验室方向是从基因组DNA变异角度系统研究肿瘤发生以及演化的规律,寻找预测肿瘤临床表现的标志物,并设计针对性的治疗策略。目前实验室的具体研究内容包括:1,从肿瘤基因组角度探讨免疫治疗临床实践中遇到的关键科学问题。如,开发新的肿瘤基因组标志物以高效区分免疫治疗有反应和不反应病人;研究肿瘤细胞免疫原性的分子遗传基础,进而设计提高肿瘤细胞免疫原性的新策略,从而把免疫治疗不反应病人变成有反应病人。2,研究肿瘤基因组DNA复杂变异的特征模式,背后驱动因素以及临床相关性。重点关注拷贝数变异(copy number alteration)、插入删除(small indel)的变异模式,与肿瘤临床尤其是免疫治疗的内在关系。3,利用生化细胞、小鼠模型研究肿瘤新靶标和机制,尤其关注新的肿瘤代谢靶点、新的非编码变异的功能与机制。实验室应用到的技术手段包括:肿瘤基因组生物信息学分析、生化细胞技术和小鼠模型。
研究成果展示
刘雪松课题组的科研工作围绕肿瘤精准诊断和治疗的实际临床需求,抓住肿瘤发生发展的根本驱动因素DNA变异,从基因组DNA变异大数据分析角度寻找肿瘤精准诊断标志物以及治疗靶点。首次报道特征性的基因组DNA变异模式(APOBEC变异模式)能作为肺癌免疫治疗效果预测的标志物;创新性地定量了肿瘤的免疫原性,并据此开发出一种比目前已知的免疫治疗标志物精确度和实用性均更为优异的标志物(肿瘤免疫原性指数,TIGS);首次发现肿瘤免疫治疗标志物TMB的临床预测效果的性别差异;开发了新的肿瘤新抗原免疫原性识别工具;首次实现肿瘤新抗原介导免疫编辑信号的系统定量;开发了一系列肿瘤基因组分析工具Sigflow, Sigminer, UCSCXenaTools, UCSCXenaShiny等;发现了新的肿瘤驱动因素与治疗靶点如GGCT,FLAD1等;上述研究成果大大扩展了对肿瘤精准诊断和治疗基因组学标志物、肿瘤新抗原疫苗开发、肿瘤新治疗靶点的认识,具有重要的实际应用场景,受到学术界的广泛关注与引用,据此也申报多项发明专利。刘雪松连续入选Elsevier发布的2021年度、2022年度“中国高被引学者”榜单。
